Chronic wasting disease (CWD) is a fatal neurodegenerative disease affecting cervids. CWD is caused by infectious prions, which can enter the environment through bodily fluids or the carcasses of infected animals. Prions can be stored, remain infectious in both soil and water for many years, and transported hydrologically, possibly expanding the geographic range of CWD transmission. In order to better predict hydrological prion transport, we investigated how CWD prion protein (PrPCWD) partitions and persists in environmental waters. We performed PrPCWD spike experiments with water samples containing fine sediments from two locations within a CWD-contaminated site, at which contamination sources were removed one year prior. Samples were filtered after spiking, and filtrates and sediments were tested separately for PrPCWD using real-time quaking-induced conversion (RT-QuIC). Unspiked filtrates tested negative for PrPCWD, while unspiked sediments were positive, indicating PrPCWD persistence in environmental sediments for at least one year. Spiked sediments were positive immediately after spiking and throughout 28 days of incubation. Spiked filtrates were largely negative immediately after spiking and remained negative for 28 days, with some inconsistent positives from one sampling location. Our results indicate that PrPCWD readily partitions to the sediment fraction of environmental waters, suggesting that hydrological prion transport is sediment-facilitated.

Environmental waters and sediments near CWD-positive animals or carcasses should be treated as potentially contaminated, even after contamination sources, such as animals and carcasses, have been removed from the area. In filtered water, our results indicate that prions can persist for at least 28 days without sediments to bind to. Additionally, unspiked sediments from both study sites exhibited positive seeding activity one year after infected deer carcasses were removed, indicating that detectable PrPCWD remained associated with sediments in the watershed for this amount of time. RT-QuIC detects prion seeding activity but does not specifically infer infectivity; therefore, it is uncertain how prion detection and movement in these environmental matrices align with potential transmission to wild cervids. One previous study shows prion infectivity persisting for over six years in both buffered solution and wastewater, (3) but prion infectivity in environmental waters remains largely understudied. Given the potential for long-term persistence of prions on environmental materials, the safety measures implemented when handling infected cervid material, including personal protective equipment and decontamination of supplies, should also apply when handling environmental materials from contaminated areas.

Detection of prions in soils contaminated by multiple routes
Results: We are able to detect prion seeding activity at multiple types of environmental hotspots, including carcass sites, contaminated captive facilities, and scrapes (i.e. urine and saliva). Differences in relative prion concentration vary depending on the nature and source of the contamination. Additionally, we have determined that prion seeding activity is retained for at least fifteen years at a contaminated site following attempted remediation.
Conclusions: Detection of prions in the environment is of the utmost importance for controlling chronic wasting disease spread. Here, we have demonstrated a viable method for detection of prions in complex environmental matrices. However, it is quite likely that this method underestimates the total infectious prion load in a contaminated sample, due to incomplete recovery of infectious prions. Further refinements are necessary for accurate quantification of prions in such samples, and to account for the intrinsic heterogeneities found in the broader environment.

Chronic wasting disease detection in environmental and biological samples from a taxidermy site
Results: The PMCA analysis demonstrated CWD seeding activity in some of the components of this facility, including insects involved in head processing, soils, and a trash dumpster.
Conclusions: Different areas of this property were used for various taxidermy procedures. We were able to detect the presence of prions in i) soils that were in contact with the heads of dead animals, ii) insects involved in the cleaning of skulls, and iii) an empty dumpster where animal carcasses were previously placed. This is the first report demonstrating that swabbing is a helpful method to screen for prion infectivity on surfaces potentially contaminated with CWD. These findings are relevant as this swabbing and amplification strategy may be used to evaluate the disease status of other free-ranging and captive settings where there is a concern for CWD transmissions, such as at feeders and water troughs with CWD-exposed properties. This approach could have substantial implications for free-ranging cervid surveillance as well as in epidemiological investigations of CWD.

This study clearly demonstrates the difficulty in removing scrapie infectivity from the farm environment. Practical and effective prion decontamination methods are still urgently required for decontamination of scrapie infectivity from farms that have had cases of scrapie and this is particularly relevant for scrapie positive goatherds, which currently have limited genetic resistance to scrapie within commercial breeds.24 This is very likely to have parallels with control efforts for CWD in cervids.

Heading Off a Wildlife Epidemic
CWD poses a significant threat to the future of hunting in Texas. Deer population declines of 45 and 50 percent have been documented in Colorado and Wyoming. A broad infection of Texas deer populations resulting in similar population impacts would inflict severe economic damage to rural communities and could negatively impact land markets. Specifically, those landowners seeking to establish a thriving herd of deer could avoid buying in areas with confirmed CWD infections. As they do with anthrax-susceptible properties, land brokers may find it advisable to inquire about the status of CWD infections on properties that they present for sale. Prospective buyers should also investigate the status of the wildlife on prospective properties. In addition, existing landowners should monitor developments as TPWD crafts management strategies to identify and contain this deadly disease.

I remember what “deep throat” told me about Scrapie back around 2001, I never forgot, and it seems it’s come to pass, from my files;

I am not sure that TSE contaminated ground could ever be free of the agent!! A very frightening revelation!!!

(I never knew who this person was, but got me into the U.S. BSE Emergency 50 State conference call back 2001, and we corresponded for years about BSE TSE Prion, have not heard from in over a few decades or so. but they were on the inside looking out. You can believe this or not, but this was real, i don’t make this stuff up…plus my endeavors to get those 1 million cattle tested for BSE failed. There was an ENHANCED BSE SURVEILLANCE put forth after 2003, we pushed for it, but it was abruptly shut down after the atypical BSE cases were popping up…a bit of history for anyone interested…terry)

The most frightening thing I have read all day is the report of Gambetti's finding of a new strain of sporadic cjd in young people...Dear God, what in the name of all that is holy is that!!! If the US has different strains of scrapie.....why???? than the UK...then would the same mechanisms that make different strains of scrapie here make different strains of BSE...if the patterns are different in sheep and mice for scrapie.....could not the BSE be different in the cattle, in the mink, in the humans.......I really think the slides or tissues and everything from these young people with the new strain of sporadic cjd should be put up to be analyzed by many, many experts in cjd........bse.....scrapie Scrape the damn slide and put it into mice.....wait.....chop up the mouse brain and and spinal cord........put into some more mice.....dammit amplify the thing and start the damned research.....This is NOT rocket science...we need to use what we know and get off our butts and move....the whining about how long everything takes.....well it takes a whole lot longer if you whine for a year and then start the research!!!
Not sure where I read this but it was a recent press release or something like that: I thought I would fall out of my chair when I read about how there was no worry about infectivity from a histopath slide or tissues because they are preserved in formic acid, or formalin or formaldehyde.....for God's sake........ Ask any pathologist in the UK what the brain tissues in the formalin looks like after a year.......it is a big fat sponge...the agent continues to eat the brain ......you can't make slides anymore because the agent has never stopped........and the old slides that are stained with Hemolysin and Eosin......they get holier and holier and degenerate and continue...what you looked at 6 months ago is not there........Gambetti better be photographing every damned thing he is looking at.....
Okay, you need to know. You don't need to pass it on as nothing will come of it and there is not a damned thing anyone can do about it. Don't even hint at it as it will be denied and laughed at.......... USDA is gonna do as little as possible until there is actually a human case in the USA of the nvcjd........if you want to move this thing along and shake the earth....then we gotta get the victims families to make sure whoever is doing the autopsy is credible, trustworthy, and a saint with the courage of Joan of Arc........I am not kidding!!!! so, unless we get a human death from EXACTLY the same form with EXACTLY the same histopath lesions as seen in the UK nvcjd........forget any action........it is ALL gonna be sporadic!!! And, if there is a case.......there is gonna be every effort to link it to international travel, international food, etc. etc. etc. etc. etc. They will go so far as to find out if a sex partner had ever traveled to the UK/europe, etc. etc. .... It is gonna be a long, lonely, dangerous twisted journey to the truth. They have all the cards, all the money, and are willing to threaten and carry out those threats....and this may be their biggest downfall...
Thanks as always for your help. (Recently had a very startling revelation from a rather senior person in government here..........knocked me out of my chair........you must keep pushing. If I was a power person....I would be demanding that there be a least a million bovine tested as soon as possible and agressively seeking this disease. The big players are coming out of the woodwork as there is money to be made!!! In short: "FIRE AT WILL"!!! for the very dumb....who's "will"! "Will be the burden to bare if there is any coverup!"
again it was said years ago and it should be taken seriously....BSE will NEVER be found in the US! As for the BSE conference call...I think you did a great service to freedom of information and making some people feign integrity...I find it scary to see that most of the "experts" are employed by the federal government or are supported on the "teat" of federal funds. A scary picture! I hope there is a confidential panel organized by the new government to really investigate this thing.
You need to watch your back........but keep picking at them.......like a buzzard to the bone...you just may get to the truth!!! (You probably have more support than you know. Too many people are afraid to show you or let anyone else know. I have heard a few things myself... you ask the questions that everyone else is too afraid to ask.)
